چکیده
Besides its hormonal effects, thyrotro pin releasing hormone (TRH) induces se veral behavioral changes in rats. Marked increases in locomotor activity occur after administration of TRH and its analogs. This effect was hypothesized to be due to a stimulation of mesolimbic dopaminergic neurons (Miyamoto and Nagawa, 1977; Heal and Green, 1979). TRH has no effect on the reuptake of monoamines (Tuomisto and Mannist, 1973) or the concentration of dopamine (DA) sensitive adenylate cyclase (Green et al. 1976). It was proposed that TRH and its analogs release DA from presyn aptic terminals of dopaminergic neurons. Kerwin and Pycock (1979) demonstrated that TRH induced 3H-DA release from the n. accumbens which was preloaded with 3H-DA. It did not release 3H-DA from the caudate nucleus. Fukuda et al. (1979) reported that unilateral injection of TRH into the nucleus caudatus pro duced rotation in rats. In biochemical

Effects of TRH and an analog, DN-1417 on the activities of single neurons in the nucleus accumbens, cerebral cortex and caudate-putamen of rats.
The actions of TRH and its analog, y butyrolactone y -carbonyl histidyl prolinamide citrate (DN-1417) on the unit activities of the cerebral cortex, caudate -putamen and nucleus accumbens were examined using a microelec trophoretic technique in anesthetized rats. Over 80% of the neurons examined in the nucleus accumbens responded to TRH and DN -1417. The effects were mainly inhibitory, but exci...
متن کاملYM-14673, a thyrotropin-releasing hormone analogue, injected into the nucleus accumbens and the striatum produces repetitive jaw movements in rats.
Bilateral injections of the thyrotropin-releasing hormone (TRH) analogue, N alpha-[((S)-4-oxo-2-azetidinyl)-carbonyl]-L-histidyl-L-prolinamide dihydrate (YM-14673, 0.1 microgram and 1 microgram/0.2 microliters), into the nucleus accumbens, the dorsal and ventrolateral striatum produced repetitive jaw movements in a dose-dependent manner. The effects were greatest in the nucleus accumbens and sm...
متن کاملThyrotropin-releasing hormone: hyperactivity and mesolimbic dopamine system in rats.
The mechanism of stimulatory action of thyrotropin-releasing hormone (TRH) on spontaneous motor activity was investigated in rats. TRH produced a significant hyperactivity with intraperitoneal administration of 20 mg/kg or bilateral injection of 10 micrograms into the nucleus accumbens septi (NAS). Following bilateral injection of 6-hydroxydopamine into the mesolimbic dopamine (DA) pathway, the...
متن کاملEffects of cannabinoids on dopamine release in the corpus striatum and the nucleus accumbens in vitro.
Cannabinoid receptors are widely distributed in the nuclei of the extrapyramidal motor and mesolimbic reward systems; their exact functions are, however, not known. The aim of the present study was to characterize the effects of cannabinoids on the electrically evoked release of endogenous dopamine in the corpus striatum and the nucleus accumbens. In rat brain slices dopamine release elicited b...
متن کاملPharmacological study of TA-0910, a new thyrotropin-releasing hormone (TRH) analog (III): Inhibition of pentobarbital anesthesia.
Sites and mechanisms of the antagonistic action of TA-0910, a new thyrotropin-releasing hormone (TRH) analog, on pentobarbital anesthesia were studied in rats. Intravenous administration of TA-0910 dose-dependently shortened the duration of pentobarbital anesthesia at 30 micrograms/kg or more. The anti-anesthetic action of TA-0910 after intracerebral injection was in the following order of effe...
